D.E.O. is a governmental organization created following Superman's arrival on Earth to monitor extra-terrestrial presence on the planet. It was formerly led by Director Hank Henshaw until he was presumably killed by Jeremiah Danvers in order to save the alien fugitive J'onn J'onzz, who began posing as Henshaw to reform the D.E.O. for its intended purpose.
After his identity was exposed, Lucy Lane was named deputy director. However, after helping Supergirl defeat Non and Indigo, J'onn received a presidential pardon and was re-instated as director of the D.E.O., continuing to use Hank's appearance for convenience sake around D.E.O. agents and for public appearances. Following Kal-El's arrival on Earth, the United States government decided that they needed to protect themselves against extra-terrestrial presence, and potential invasion, so they created the Department of Extra-Normal Operations.

The Central City Police Department (CCPD) is a police department servicing Central City, CCPD has both ally and opponent of Flash, the superhero long-established in Central City.
24 years following, after Fort Rozz crashed on Earth, they began to catalog of all of the aliens seen in public. Twelve years later, they captured Kara Danvers. She was made aware of the department's existence, and their motives. Though they initially didn't work well, Hank Henshaw allowed Kara to take down Vartox, an alien threat, as Supergirl.

- J'onn J'onzz / Martian Manhunter : Head of the DEO who takes Hank Henshaw's likeness after Henshaw is killed in Peru while hunting J'onn. J'onn takes Henshaw's likeness in order to reform the DEO from within as well as to watch over Alex and Kara.
- Alex Danvers : She is a scientist and government agent who works for Hank Henshaw at the DEO, serving as his right-hand. Having been extensively trained in combat after joining the DEO, Alex in turn provided rigorous training to Kara in order to decrease her reliance on her powers.
- Kara Danvers / Kara Zor-El / Supergirl : Kryptonian living in National City, who must embrace her powers after previously hiding them. She assists her adoptive sister as part of the Department of Extra-Normal Operations as she discovered the truth that her adoptive father also worked for the DEO so they would not take her, while Alex's co-workers at the DEO help her perfect her powers.
- Winn Schott : A tech expert who worked alongside Kara at CatCo, he is Kara's best friend and serves as one of her allies, helping her develop her costume and aiding her in her adventures. Winn has unrequited feelings for Kara and is a rival with James for her affection.
- Vasquez : A DEO agent who often works closely with Hank (J'onn), Alex, and Kara/Supergirl and is responsible for coordinating/relaying information on alien attacks or other incidents.
